PCR DNA and Gel Band Purification Kit, GE Healthcare
Kits are designed for the rapid purification and concentration of PCR products and DNA fragments ranging in size from 50bp–10kb. Can be used to
purify DNA from reaction volumes up to 100μL or agarose gel slices up to 900mg. The kits combine a versatile chaotropic buffer with a glass fiber
matrix supported in a spin-column. Typical recoveries range from 60–80% for DNA fragments from agarose gels and as high as 95% for PCR prod-
ucts from solution. DNA purity is exceptional; 99.5% of contaminants are removed.

Kit includes GFX columns, collection tubes and color-coded bottles of capture buffer, wash buffer, and two elution buffers
(Tris-HCl and sterile water).

Specialty Vectors, Lucigen
The pJW168 vector is designed for in vivo insertion/removal of a DNA sequence through cre-lox site-specific recombination. The vector con-
tains the cre-recombinase gene under the control of the IPTG-inducible lacUV5 promoter, as well as unique restriction sites for cloning and the
gene for ampicillin selection. This vector allows DNA fragments to be integrated and then completely removed from the bacterial chromosome. Following recombina-
tion, growing transformed bacterial cultures at 42°C (108°F) will efficiently eliminate pJW168 and cease the production of cre recombinase.
The plnvRecA vector is designed to enable P1 transduction in any E.coli strain, making it highly useful in generating mutant strains for microbial genomic and pro-
teomic studies, or for moving genes from one strain to another. This vector can be used in any E. coli strain, unlike other P1 transduction strategies, which can only
be used with wild-type (RecA+) E. coli strains. The plnvRecA vector transiently converts RecA– to RecA+. After transduction, the RecA– phenotype can be completely
restored by growing the cells at 42°C (108°F).
The pRANGER
-BTB vectors provide ultra-high stability cloning in a broad host-range expression vector. They incorporate the pBBR1 replicon to enable stable plasmid
replication in Gram-negative bacteria. They also contain the pBAD promoter for inducible expression of cloned genes with a simple arabinose incubation. These fea-
tures allow cloning and expression in many Gram-negative strains and avoid the structural and segregational instability that may occur with standard E. coli cloning
vectors. pRANGER-BTB vectors are available in a choice of four antibiotic resistance cassettes.
The pSMART
®
cDNA-SS4 vector enables accurate and efficient production of single-stranded cDNAs for libraries, probes, and microarrays. The ligation-ready vector in-
corporates CloneSmart
®
technology for maximum insert stability with very low background, as well as the f1 origin for single-stranded DNA production, a T7 promoter
for in vitro or in vivo expression, and convenient blunt+NotI cloning sites. The vector's cloning efficiency is greater than 99%, virtually eliminating the need for colony
screening. It is provided dephosphorylated and precut at blunt-NotI sites in a 4X vector premix containing buffer and ATP.
The pTrueBlue
®
vector eliminates common problems in blue/white screening procedures by positioning the cloning sites within essential coding sequences that do not
permit the formation of active [beta]-Gal fusion proteins. With the use of this vector, blue colonies can not be recombinants. The vector features 31 restriction sites for
cloning, 8-base sites flanking the multiple clone site for clone mapping, ExoIII-protection sites for creation of unidirectional nested deletions, an f1 origin for ssDNA
production, and a T7 promoter for making RNA probes. The vector has a high copy number (200–500 per cell) and can be used to clone inserts up to 3kb.
All vectors are provided as a 500μL E. coli glycerol stock containing the intact plasmid.
